Nanoscale Amperometry Reveals that Only a Fraction of Vesicular Serotonin Content is Released During Exocytosis from Beta Cells

چکیده
Recent work has shown that chemical release during the fundamental cellular process of exocytosis in model cell lines is not all-or-none. We tested this theory for vesicular from single pancreatic beta cells. The vesicles these cells insulin, but also serotonin, which detectible with amperometric methods. Traditionally, it assumed Here, we use a multidisciplinary approach involving nanoscale methods to explore nature insulin exocytosis. amperometrically quantified number serotonin molecules stored inside individual (39 317±1611) cytoplasm before and released (13 310±1127) each stimulated event. Actin Controls the Vesicular Fraction of Dopamine Released During Extended Kiss and Run Exocytosis
The effect of latrunculin A, an inhibitor of actin cross-linking, on exocytosis in PC12 cells was investigated with single cell amperometry. This analysis strongly suggests that the actin cytoskeleton might be involved in regulating exocytosis, especially by mediating the constriction of the pore. In an extended kiss-and-run release mode, actin could actually control the fraction of neurotransm...
متن کاملDetection of exocytosis at individual pancreatic beta cells by amperometry at a chemically modified microelectrode.
Amperometry at a carbon fiber microelectrode modified with a composite of ruthenium oxide and cyanoruthenate was used to monitor chemical secretions of single pancreatic beta cells from rats and humans. When the insulin secretagogues glucose, tolbutamide, and K+ were applied to the cell, a series of randomly occurring current spikes was observed. The current spikes were shown to be due to the d...
متن کاملEvidence that an isoform of calpain-10 is a regulator of exocytosis in pancreatic beta-cells.
Calpain-10 (CAPN10) is the first type 2 diabetes susceptibility gene to be identified through a genome scan, with polymorphisms being associated with altered CAPN10 expression. Functional data have been hitherto elusive, but we report here a corresponding increase between CAPN10 expression level and regulated insulin secretion. Pancreatic beta-cell secretory granule exocytosis is mediated by th...
متن کامل'Full fusion' is not ineluctable during vesicular exocytosis of neurotransmitters by endocrine cells.
Vesicular exocytosis is an essential and ubiquitous process in neurons and endocrine cells by which neurotransmitters are released in synaptic clefts or extracellular fluids. It involves the fusion of a vesicle loaded with chemical messengers with the cell membrane through a nanometric fusion pore. In endocrine cells, unless it closes after some flickering ('Kiss-and-Run' events), this initial ...
متن کاملEvidence that release of adenosine triphosphate from endothelial cells during increased shear stress is vesicular.
In response to increased shear stress, vascular endothelial cells release adenosine triphosphate (ATP) by an unknown mechanism. We have investigated this mechanism using different approaches. First, we discovered that quinacrine, used to locate intracellular stores of ATP bound to peptides, displayed a granular fluorescence, typical of vesicular storage. Second, we found that two inhibitors of ...
